What is Tadalafil?

Class and mechanism: Tadalafil is classified as potent and highly selective phosphodiesterase type 5 (PDE5) inhibitors. Inhibition of PDE5 prevents cyclic guanosine monophosphate (cGMP) from being degraded in penile tissue. When the enzyme is inhibited, relaxation of the smooth muscle in the corpus cavernosum occurs and leads to inflow of blood, potentiating erection. The drug requires sexual stimulation for response (Palacioz, 2003; Thomson Micromedex, 2003).
Tadalafil Storage
Keep this medication in the container it came in, tightly closed, and out of reach of children. Store it at room temperature and away from excess heat and moisture (not in the bathroom). Throw away any medication that is outdated or no longer needed. Talk to your pharmacist about the proper disposal of your medication.
How to take Tadalafil?
Tadalafil comes as a tablet to take by mouth. It is usually taken with or without food before sexual activity. Your doctor will help you decide the best time for you to take Tadalafil before sexual activity. Tadalafil should not be taken more often than once every 24 hours. If you have certain health conditions or are taking certain medications, your doctor may tell you to take Tadalafil less often. Follow the directions on your prescription label carefully, and ask your doctor or pharmacist to explain any part you do not understand. Take Tadalafil exactly as directed. Do not take more or less of it or take it more often than prescribed by your doctor.
What to do if I miss a dose?
Tadalafil is taken as needed and there is no missed dose that is likely to happen.
The symptoms of Tadalafil OVERDOSE are not yet known but if Tadalafil overdose is suspected, you have to seek medical attention immediately.
Who should not take Tadalafil?
Tadalafil should not be used by patients taking nitrates (such as nitroglycerin tablets or patches) or any alpha blocker other than FLOMAX 0.4mg daily because the combination of these with Tadalafil could significantly lower blood pressure and lead to fainting or even death in some men.
What side effects may occur?
As with any drug product, there may be side effects. The most commonly reported side effects in patients treated with Tadalafil during the testing of the product were:
- headache
- indigestion
- back pain
- muscle pain
- flushing
- stuffy nose
Patients who get back pain and muscle aches usually get them 12 to 24 hours after taking Tadalafil and these usually go away by themselves within 48 hours. A small number of patients taking Tadalafil also reported abnormal vision.
Tadalafil Drug Interactions
Do not take Tadalafil together with the following medications as they may cause very low blood pressure and may cause extreme dizziness, fainting, heart attack and even stroke.
- Alpha blockers such as doxazosin
- Nitrates
- Nitroglycerin drugs
Tell your doctor if you are taking other drugs to treat erectile dysfunction such as Viagra, Vardenafil, Yocon and others.
You doctor may advise you not to take Tadalafil or prescribe a much lower dosage for you of you are taking any of the drugs mentioned.
Additional Information
Tell your doctor about your medical conditions and all medications, and ask if you're healthy enough for sexual activity. Don't take Tadalafil if you take nitrates, often prescribed for chest pain, as this may cause a sudden, unsafe drop in blood pressure. Don't drink alcohol in excess (to a level of intoxication) with Tadalafil, as this may increase your chances of getting dizzy or lowering your blood pressure.
Sexual stimulation is required for Tadalafil to work. You will not get an erection just by taking Tadalafil without sexual stimulation.
